Gene Expression and Discovery During Lens Regeneration in Mouse: Regulation of Epithelial to Mesenchymal Transition and Lens Differentiation

    Get PDF
Purpose: It has been shown that after extracapsular lens removal by anterior capsulotomy in the mouse, the lens can be regenerated. However, as the capsular bag is filled with fibers, epithelial to mesenchymal transition (EMT), an event which is common after cataract surgery as well, takes place during early stages. This study, using a unique mouse model, was undertaken to identify novel regulators and networks in order to more clearly understand secondary cataracts at the molecular level

The analysis uses a data sample of pp collisions corresponding to an integrated luminosity of 5 fb-1 collected by the CMS experiment at the LHC at √s=7 TeV in 2011. No significant excess is observed in the mass distribution of the VZ candidates compared with the background expectation from standard model processes. Model-dependent upper limits at the 95% confidence level are set on the product of the cross section times the branching fraction of hypothetical particles decaying to the VZ final state as a function of mass. Sequential standard model W′ bosons with masses between 700 and 940 GeV are excluded. In the Randall-Sundrum model for graviton resonances with a coupling parameter of 0.05, masses between 750 and 880 GeV are also exclude

Discutindo a educação ambiental no cotidiano escolar: desenvolvimento de projetos na escola formação inicial e continuada de professores

    Get PDF

Letter to the Editor Lens regeneration in mice: implications in cataracts

Abstract Lens regeneration in adult mice is possible when the lens capsule is left behind after lentectomy. The lens is regenerated by the remaining adherent lens epithelial cells, which differentiate to form lens fibres within days, showing normal morphology and bow regions. Epithelial to mesenchymal cell transformation is also seen during the early stages. The mouse, therefore, can become an indispensable animal model for cataract research, surgery and therapy. q 2003 Elsevier Ltd. All rights reserved. Keywords: lens; regeneration; mouse; cataract Traditionally, the newt has been hailed as the most powerful animal model for lens regeneration We used three different strains in our study, Balb/c, NZW and MRL/MpJ þ/þ . The mice were sexually mature (8 -12 weeks old) of both sexes. Before operation, mice were anesthetized with ketamine (87 mg kg 21 ) in combination with xylazine (13 mg kg 21 ). Two types of operations were performed. In one set, the lens along with the capsule was removed and in the other set, the capsule was left behind (with only part of the anterior capsule damaged). Integrity and restoration of the normal shape of the capsule is important. To remove the lens, a corneal incision was made with a sharp blade. Due to the small size of the mouse eye, we preferred to remove the lens with fine forceps by applying pressure in the eye. Such an operation results, as in the newts, in removal of the lens and the capsule. However, if an incision is made in the anterior lens as well this operation leads to removal of the lens but not the capsule. After such extracapsular extraction, we injected saline solution to clean the capsule. Histological preparations of eyes after lentectomy showed a rather clean capsule with adherent lens epithelial cells. After lentectomy, the mice were collected in time intervals starting at 2 days and ending at 30 days post lentectomy. We found that when the whole lens (with the capsule) was removed no lens regeneration resulted in any cas

Mouse gene targeting reveals an essential role of mTOR in hematopoietic stem cell engraftment and hematopoiesis

mTOR integrates signals from nutrients and growth factors to control protein synthesis, cell growth, and survival. Although mTOR has been established as a therapeutic target in hematologic malignancies, its physiological role in regulating hematopoiesis remains unclear. Here we show that conditional gene targeting of mTOR causes bone marrow failure and defects in multi-lineage hematopoiesis including myelopoiesis, erythropoiesis, thrombopoiesis, and lymphopoiesis. mTOR deficiency results in loss of quiescence of hematopoietic stem cells, leading to a transient increase but long-term exhaustion and defective engraftment of hematopoietic stem cells in lethally irradiated recipient mice. Furthermore, ablation of mTOR causes increased apoptosis in lineage-committed blood cells but not hematopoietic stem cells, indicating a differentiation stage-specific function. These results demonstrate that mTOR is essential for hematopoietic stem cell engraftment and multi-lineage hematopoiesis

Gene expression signatures in the newt irises during lens regeneration

AbstractLens regeneration in adult newts is possible by transdifferentiation of the pigment epithelial cells (PECs) of the dorsal iris. The same cells in the ventral iris are not capable of such a process. To understand this difference in regenerative competency, we examined gene expression of 373 genes in the intact dorsal and ventral irises as well as in irises during the process of lens regeneration. We found similar signatures of gene expression in dorsal and ventral with several cases of even higher levels in the ventral iris. Such transcriptional activity in the regeneration-incompetent ventral iris was unexpected and calls for a revision of our views about mechanisms of lens regeneration induction
